Relationship between Histoltgic Change and Expression of COX-2 on CCl©þ-Induced Cirrhosis in the Rat

Abstract
COX-2 is an inducible immediate-early gene and was orginally found to be induced by various stimuli, such as mitogens and cytokines, and growth factors. Overexpression of COX-2 has been reported in various chronic inflammatory disease. The aim of this work was to examine the relationship between the expression of COX-2 and developement of the CCl4-induced cirrhosis. The rats were sacrificed at 2, 4, 6, 8, and 12 experimental weeks. The light microscopic, immunohistochemical, and TUNEL stain, along with the liver function tests were performed. The number of Kupffer cells and endothelial cell showing the expression of COX-2 and the apoptosis of hepatocytes were gradually increased during the development of cirrhosis. In the 12 weeks, the hepatocytes also showed the expression of COX-2. In serologic test, the level of total protein and albumin decreased, but the degree of aspartate aminotransferase, alanine aminotransferase, and alkaline phosphatase levels increased and the levels of increase are proportional to the experimental duration. The COX-2 expression showed the correlationship between the degree of fibrosis and the liver function test. So, it suggests that COX-2 participate in development of the liver cirrhosis and the selective inhibition of the COX-2 may prevent the progression of cirrhosis with its complication including the hepatocellular carcinoma.
